- Título: DYSLIPIDEMIC APOCIII TRANSGENIC MICE DO NOT PRESENT CHANGES RELATED TO THE NUMBER OF INTRAEPITHELIAL LYMPHOCYTES (IEL’S) IN THE DUODENUM

Introduction: Mice that overexpress human apolipoprotein CIII (apoC III) have high levels of triglycerides and are considered dyslipidemic. This protein is mainly synthesized in the liver, muscle and to a fewer amount in the intestine and is involved in inflammatory processes and regulation of the complement system. The intraepithelial lymphocytes (IEL’s) are in the intestinal epithelium, these cells migrate from the lamina propria as defense mechanism of the intestinal immune barrier, reacting quickly to stimuli from diet or microbiota. Objective: The objective of this study was to quantify IEL’s present in the duodenum of mice that overexpress human apoCIII and non-transgenic littermates control mice. Methods: This study was approved by the Committee of Ethics in Research in Animal Experiments of the State University of Maringá with registration nº 020/2013 and by the Internal Commission of Biosafety nº 819/2013.Twelve C57BL/6 mice were randomly divided into two groups (n=6): transgenic animals (Tg) and non-transgenic controls animals (Ntg). After euthanasia the duodenum was collected and submitted to the histological process to obtain semi-serial sections of 4μm stained by the HE technique. The number of IEL's present between 2560 epithelial cells per mouse was counted and then the ratio of to 100 epithelial cellswas calculated. Statistical analysis was performed by Bioestat 5.0 software, using the Shapiro-Wilk normality test followed by the T-student test by Grand Pad Prism 5.01 software (p<0.05). Results: There wasn’t significant change in the number of IEL’s (13.47 ± 2.40IEL's / 100 epithelial cells) in the TG when compared to the Ntg (12.34 ± 1.72 IEL's / 100 epithelial cells). Conclusion: Our data show that overexpression of human ApoCIII does not interfere in the counting of IEL's in the duodenum of C57BL/6 mice.
